    Enzymatic hydrolysate from velvet antler suppresses adipogenesis in 3T3-L1 cells and attenuates obesity in high-fat diet-fed mice

    The purpose of the current study was to investigate the potential anti-obesity activity of an enzymatic hydrolysate of velvet antler in inhibiting adipogenesis in 3T3-L1 cells and in high-fat diet (HFD)-fed obese mice. The enzy- matic hydrolysate was prepared using the commercial food grade protease, Protamex. The velvet antler Protamex hydrolysate (VAPH) indicated profound inhibitory effects on adipogenesis dose-dependently by decreasing the accumulation of triglycerides and down-regulating expression levels of adipogenesis-related proteins C/EBPα, SREBP-1, and PPARγ. In a mouse model of HFD-induced obesity, oral administration of VAPH (100 and 300 mg/kg for 13 weeks) significantly reduced the body weight gain that had resulted from the HFD. VAPH treat- ment also lowered the serum glucose and triglyceride levels, while increasing the HDL-C level. Furthermore, the treatment greatly reduced hepatic lipid droplet accumulation as well as the size of adipocytes. Current findings H has profound anti-obesity effects and could be an effective candidate for preventing obesity and obesity-related chronic diseases

    Modeling study of human renal chloride channel (hCLC-5) mutations suggests a structural-functional relationship.BackgroundDent's disease, a renal tubular disorder characterized by low-molecular-weight proteinuria, hypercalciuria, and nephrolithiasis, is due to inactivating mutations in the X-linked renal-specific chloride channel, hCLC-5. The x-ray crystal structures of two bacterial chloride channels (CLCs) have recently been established, thereby allowing us to construct a model for hCLC-5 and further examine the role of its mutations.MethodsThe data regarding 49 hCLC-5 mutations were reviewed. Thirty-four mutations that predicted absent or truncated channels were excluded. The remaining 15 mutations (one in-frame insertion and 14 missense mutations), 12 of which have been studied electrophysiologically, were assessed. The hCLC-5 sequence was aligned with the Salmonella typhimurium and Escherichia coli sequences and used to map the hCLC-5 mutations onto a three-dimensional model.ResultshCLC-5 is a homodimeric protein, with each subunit consisting of 18 helices. None of the missense mutations involved the chloride (Cl−) selectivity filter, but 12 of the 15 mutations were found to be clustered at the interface of the two subunits. Six of these mutations occurred in two of the helices that either form part of the interface or lie in close proximity to the interface, and three other mutations that did not lead to complete loss of Cl− conductance were at the edge of the interface.ConclusionThese results demonstrate a crucial role for the interaction between the two subunits at the interface of the homodimeric hCLC-5

This is an NP-hard problem. Several approach methods have been developed to solve this problem. In this paper, we propose a new encoding scheme which is used in a hybrid resolution: a metaheuristic is matched with a list algorithm (Next Fit, First Fit, Best Fit) to solve the bin packing problem. Any metaheuristic can be used but in this paper, our proposition is implemented on a single solution based metaheuristic (stochastic descent, simulated annealing, kangaroo algorithm). This hybrid method is tested on literature instances to ensure its good results

    Background-Arrhythmogenic Right Ventricular Dysplasia/Cardiomyopathy (ARVD/C) Diagnostic Task Force Criteria (TFC) proposed in 1994 are highly specific but lack sensitivity. A new international task force modified criteria to improve diagnostic yield. A comparison of diagnosis by 1994 TFC versus newly proposed criteria in 3 patient groups was conducted. Methods and Results-In new TFC, scoring by major and minor criteria is maintained. Structural abnormalities are quantified and TFC highly specific for ARVD/C upgraded to major. Furthermore, new criteria are added: terminal activation duration of QRS ≥55 ms, ventricular tachycardia with left bundle-branch block morphology and superior axis, and genetic criteria. Three groups were studied: (1) 105 patients with proven ARVD/C according to 1994 TFC, (2) 89 of their family members, and (3) 39 patients with probable ARVD/C (ie, 3 points by 1994 TFC). All were screened for pathogenic mutations in desmosomal genes. Three ARVD/C patients did not meet the new sharpened criteria on structural abnormalities and thereby did not fulfill new TFC. In 62 of 105 patients with proven ARVD/C, mutations were found: 58 in the gene encoding Plakophilin2 (PKP2), 3 in Desmoglein2, 3 in Desmocollin2, and 1 in Desmoplakin.     An Improved Upper Bound for the Ground State Energy of Fermion Lattice Models

    We present an improved upper bound for the ground state energy of lattice fermion models with sign problem. The bound can be computed by numerical simulation of a recently proposed family of deformed Hamiltonians with no sign problem. For one dimensional models, we expect the bound to be particularly effective and practical extrapolation procedures are discussed. In particular, in a model of spinless interacting fermions and in the Hubbard model at various filling and Coulomb repulsion we show how such techniques can estimate ground state energies and correlation function with great accuracy.Comment: 5 pages, 5 figures; to appear in Physical Review

    Poly(vinylidene fluoride) and copolymers as porous membranes for tissue engineering applications

    Poly(vinylidene fluoride) (PVDF) and its main copolymers - poly(vinylidene fluoride-co-hexafluoropropene), P(VDF-HFP), and poly(vinylidene fluoride-co-trifluoroethylene), P(VDF-TrFE) - were processed by solvent casting at room temperature in the form of porous membranes. Copolymer membranes showed higher degree of porosity than PVDF, the average pore size being larger for P(VDF-TrFE) than for P(VDF-HFP) and PVDF. All membranes show high hydrophobicity with water contact angles in the range 94° to 115°, and electroactive beta phase contents above 90%. The adhesion and proliferation of both C2C12 myoblast and MC3T3-E1 pre-osteoblast cells on the membranes were investigated. It is demonstrated that PVDF membranes promote higher cell proliferation while P(VDF-HFP) membranes show the lowest proliferation for both kinds of cell. The proliferation on P(VDF-TrFE) membranes is cell dependent, higher for MC3T3-E1 cells but lower for C2C12 cells, related to the effect of the highly porous structure on the preferred morphology of each cell type, as the higher pore size and porosity of the P(VDF-TrFE) membrane induce cell elongation, which is preferred just by the C2C12 muscle cells.Funded by FEDER funds through the “Programa Operacional Fatores de Competitividade e COMPETE” and by national funds arranged by FCT Fundação para a Ciência e a Tecnologia, project references PTDC/CTM-NAN/112574/2009 and PEST-C/FIS/UI607/2014.     Measurements of long-range near-side angular correlations in sNN=5\sqrt{s_{\text{NN}}}=5TeV proton-lead collisions in the forward region

    Two-particle angular correlations are studied in proton-lead collisions at a nucleon-nucleon centre-of-mass energy of sNN=5\sqrt{s_{\text{NN}}}=5TeV, collected with the LHCb detector at the LHC. The analysis is based on data recorded in two beam configurations, in which either the direction of the proton or that of the lead ion is analysed. The correlations are measured in the laboratory system as a function of relative pseudorapidity, Δη\Delta\eta, and relative azimuthal angle, Δϕ\Delta\phi, for events in different classes of event activity and for different bins of particle transverse momentum. In high-activity events a long-range correlation on the near side, Δϕ0\Delta\phi \approx 0, is observed in the pseudorapidity range 2.0<η<4.92.0<\eta<4.9. This measurement of long-range correlations on the near side in proton-lead collisions extends previous observations into the forward region up to η=4.9\eta=4.9. The correlation increases with growing event activity and is found to be more pronounced in the direction of the lead beam.     Study of the production of Λb0\Lambda_b^0 and B0\overline{B}^0 hadrons in pppp collisions and first measurement of the Λb0J/ψpK\Lambda_b^0\rightarrow J/\psi pK^- branching fraction

    The product of the Λb0\Lambda_b^0 (B0\overline{B}^0) differential production cross-section and the branching fraction of the decay Λb0J/ψpK\Lambda_b^0\rightarrow J/\psi pK^- (B0J/ψK(892)0\overline{B}^0\rightarrow J/\psi\overline{K}^*(892)^0) is measured as a function of the beauty hadron transverse momentum, pTp_{\rm T}, and rapidity, yy. The kinematic region of the measurements is pT<20 GeV/cp_{\rm T}<20~{\rm GeV}/c and 2.0<y<4.52.0<y<4.5. The measurements use a data sample corresponding to an integrated luminosity of 3 fb13~{\rm fb}^{-1} collected by the LHCb detector in pppp collisions at centre-of-mass energies s=7 TeV\sqrt{s}=7~{\rm TeV} in 2011 and s=8 TeV\sqrt{s}=8~{\rm TeV} in 2012. Based on previous LHCb results of the fragmentation fraction ratio, fΛB0/fdf_{\Lambda_B^0}/f_d, the branching fraction of the decay Λb0J/ψpK\Lambda_b^0\rightarrow J/\psi pK^- is measured to be \begin{equation*} \mathcal{B}(\Lambda_b^0\rightarrow J/\psi pK^-)= (3.17\pm0.04\pm0.07\pm0.34^{+0.45}_{-0.28})\times10^{-4}, \end{equation*} where the first uncertainty is statistical, the second is systematic, the third is due to the uncertainty on the branching fraction of the decay B0J/ψK(892)0\overline{B}^0\rightarrow J/\psi\overline{K}^*(892)^0, and the fourth is due to the knowledge of fΛb0/fdf_{\Lambda_b^0}/f_d. The sum of the asymmetries in the production and decay between Λb0\Lambda_b^0 and Λb0\overline{\Lambda}_b^0 is also measured as a function of pTp_{\rm T} and yy. The previously published branching fraction of Λb0J/ψpπ\Lambda_b^0\rightarrow J/\psi p\pi^-, relative to that of Λb0J/ψpK\Lambda_b^0\rightarrow J/\psi pK^-, is updated. The branching fractions of Λb0Pc+(J/ψp)K\Lambda_b^0\rightarrow P_c^+(\rightarrow J/\psi p)K^- are determined.Comment: 29 pages, 19figures.     Evidence for the strangeness-changing weak decay ΞbΛb0π\Xi_b^-\to\Lambda_b^0\pi^-

    Using a pppp collision data sample corresponding to an integrated luminosity of 3.0~fb1^{-1}, collected by the LHCb detector, we present the first search for the strangeness-changing weak decay ΞbΛb0π\Xi_b^-\to\Lambda_b^0\pi^-. No bb hadron decay of this type has been seen before. A signal for this decay, corresponding to a significance of 3.2 standard deviations, is reported. The relative rate is measured to be fΞbfΛb0B(ΞbΛb0π)=(5.7±1.80.9+0.8)×104{{f_{\Xi_b^-}}\over{f_{\Lambda_b^0}}}{\cal{B}}(\Xi_b^-\to\Lambda_b^0\pi^-) = (5.7\pm1.8^{+0.8}_{-0.9})\times10^{-4}, where fΞbf_{\Xi_b^-} and fΛb0f_{\Lambda_b^0} are the bΞbb\to\Xi_b^- and bΛb0b\to\Lambda_b^0 fragmentation fractions, and B(ΞbΛb0π){\cal{B}}(\Xi_b^-\to\Lambda_b^0\pi^-) is the branching fraction. Assuming fΞb/fΛb0f_{\Xi_b^-}/f_{\Lambda_b^0} is bounded between 0.1 and 0.3, the branching fraction B(ΞbΛb0π){\cal{B}}(\Xi_b^-\to\Lambda_b^0\pi^-) would lie in the range from (0.57±0.21)%(0.57\pm0.21)\% to (0.19±0.07)%(0.19\pm0.07)\%.Comment: 7 pages, 2 figures, All figures and tables, along with any supplementary material and additional information, are available at https://lhcbproject.web.cern.ch/lhcbproject/Publications/LHCbProjectPublic/LHCb-PAPER-2015-047.htm

    BB flavour tagging using charm decays at the LHCb experiment

    An algorithm is described for tagging the flavour content at production of neutral BB mesons in the LHCb experiment. The algorithm exploits the correlation of the flavour of a BB meson with the charge of a reconstructed secondary charm hadron from the decay of the other bb hadron produced in the proton-proton collision. Charm hadron candidates are identified in a number of fully or partially reconstructed Cabibbo-favoured decay modes. The algorithm is calibrated on the self-tagged decay modes B+J/ψK+B^+ \to J/\psi \, K^+ and B0J/ψK0B^0 \to J/\psi \, K^{*0} using 3.0fb13.0\mathrm{\,fb}^{-1} of data collected by the LHCb experiment at pppp centre-of-mass energies of 7TeV7\mathrm{\,TeV} and 8TeV8\mathrm{\,TeV}.
